The SHH/SMO pathway maintains OPCs in an undifferentiated state but is not essential for their final maturation or myelin formation. Experimental models suggest that modulating this pathway could enhance remyelination in diseases like multiple sclerosis and leukodystrophies. However, precise regulation is necessary to avoid adverse effects, such as tumorigenesis. The article highlights both canonical and non-canonical SHH signaling mechanisms and emphasizes the therapeutic potential of SHH/SMO “switches” to control OPC proliferation and differentiation.
